Abstract

ABSTRACT

Fish Quarantine Installation (FQI) is a building or room with equipment, land, and other supporting facilities needed as a place to carry out isolation actions. Calcium hypochlorite (Ca(ClO)2) or chlorine known to the general public as kaporit in Indonesia. Chlorine is a disinfectant that is widely used in Fish Farming Business Units for sterilization of facilities (equipment) in fish quarantine installations. The dosage and duration of use of chlorine disinfectants still vary. The aims of this study are to obtain an effective dose and immersion time of chlorine disinfectant. Aeromonas salmonicida is bacteria commonly attack freshwater ornamental fish and consumption fish. The bacteria was used for contamination of FQI equipment with a density of 10 8 cfu/ml. The FQI equipment used was aeration stone, scoop, filter cotton, and Styrofoam. FQI equipment was contaminated with bacteria by soaking/dipping. FQI equipment contaminated with bacteria was disinfected with chlorine doses of 0 ppp (negative control), 25 ppm, 50 ppm, 75 ppm, 100 ppm, 125 ppm, 150 ppm, and positive control (commercial disinfectant) with a soaking time of 1 hour, 3 jams, 6 hours and 12 hours. Bacteria were isolated from the treated equipment. The results of the observation showed the growth of bacterial colonies in the negative control (0 ppm) from a soaking time of 1 hour to 12 hours, at a dose of 25 ppm bacterial colonies grew at a soaking time of 1 hour and 3 hours, while at doses of 50 ppm, 75 ppm, 100 ppm, 125 ppm, 150 ppm and positive control there was no growth of bacterial colonies either at a soaking time of 1 hour, 3 hours, 6 hours and 12 hours. Effective chlorine for disinfecting live freshwater fish quarantine installation equipment is at a dose of 50 ppm. Meanwhile, the effective soaking time for chlorine disinfectant is 1 hour at a chlorine dose of 50 ppm in a closed place/container.
